I have my Instrument, Commercial, Multi-Engine,CFI, MEI, CFII, and have failed 2 checkrides. Is this a big deal to employers for airline jobs?

It's not good to have failed twice but being hired as a pilot for airlines and for UPS and FedEx is more about who you know. There are so many applications to airlines and freight companies by pilots that the only way to get noticed is if someone working there personally hands in your resume to the hiring people.
Also make sure you have not gotten in trouble with the law. Even misdemeanors will be questioned during the interview.

Unfortunately yes. Air carriers are required to contact previous employers regarding your performance as a pilot. Some carriers will have to terminate you if you fail a check-ride twice during your employment.
Also DUI convictions are very tough for a pilot, you need to disclose at the time of your medical an in any job interview.

No. I failed my private and no airline seemed to care. I was offered three jobs and turned them all down.
Then again, if you fail EVERY checkride....
